Welcome to the forum CM, Garrison Outfitters make a kit for the rubber:

Garrison Outfitters (http://garrisonoutfitters.com/product-catalogue/vehicle-equipment/window-channel-kits/rubber-window-channel-for-land-rover-defender-rear-sliding-window.html)

It's a bit pricey and hard to get, but at least you'll know it works first go.

I took the window out, took the frame down to my local glazier, who fitted a piece of glass (same stuff windscreens are made of - cant remember what its called) using silicone. I brought it home and re-rivetted it in place. I cant remember exactly how mich it cost, but it was less than $100, and maybe half that.

Welcome , I agree with towe , talk to a couple of your local glass company's , tell them the size , and ask how much to cut you a bit of Laminate glass . There are a few post's about fitting side windows , or Gull windows , that should give you a bit of idea how they come out !!.. Jm ..
