The Wall at Derwent Bridge if you appreciate remarkable craftsmanship.
The Ida Bay Railway if you are interested in the history of that part of Tas.
Cockle Creek for a camp and the satisfaction of being as far south as you can drive in Australia.
    The Lavender Farm at Bridport.
Tin Dragon Interpretation Centre in Derby
Waterhouse Conservation area is a great spot to camp.
    Ranga Flat.
do the lavender farm right at the end if you want to see the flowers. They start harvest at Christmas.

All the the other suggestion in this and other posts are good

Pub in the paddock leads to the Pyengana Jeep Track(starts off quite innocently before becoming quite challenging!). You can Pitch a tent in the paddock adjacent to the pub. Great food and draught beer,Mmmmmmmmmmm.
The Wall is an awesome display of what a craftsman can do with a chisel!
If you are going to Cradle Mountain then try and book at the Waldheim Cabins. By doing that you stay within the park and the place turns into Wombat city when the day trippers leave, also the story of how they( Weindorfers) established Cradle Mountain as a park for all to enjoy is a testament of true grit and determination!
Montezuma Falls is a fun 4WD track and the suspension bridge takes your breath away!
If you have time we do the ‘Three Capes Walk’ it’s the best hike I’ve done in Australia.
Port Arthur’s Ghost tour(in the evening) is great.
Scallop Pies,Scallop Pies,Scallop Pies,...........OMG, how good are the Scallop Pies. Get one into you!
The Nut at Stanley was also specky. You can also watch Fairy Penguins on the local beach as the sun sets.
MONA was awesome, kids loved the ‘Poo machine’!
Lavender farm was very pretty, if you join their club online admission is free. However also make sure they haven’t harvested as if there are no flowers it’s not pretty at all.
    Enjoy!!

Mt Mcall track to Franklin River, near Queenstown has to be one of the most scenic tracks/ drives in Tasmania. Seeing as you need to get a key from parks in advance, you are guaranteed to have it to yourself too. Another key required track is the east west trail on mt Wellington. These kinds of experiences are good to add to a Tassie trip because they're a bit different to the usual tourist stuff. (You should do all the tourist stuff too tho!)

Have a few videos of most locations mentioned here, the drive up to Lake Cumberland is worth it if you have a fairly modified vehicle (probably hardest technical track in Tassy)
