In the first example, there are 4 entities whom he has thanked - his parents, God, and Mrs. Trunchbull.

In the second example, the missing comma makes the sentence read such that his parents are God and Mrs Trunchbull. The parents are no longer separate entities.

Read it aloud but making a distinct pause at each comma and you'll hear the difference.
