I had a misfire in my TD5 Defender. It was very intermittent at first, but when it was misbehaving, it felt very much like a petrol engine with a crook plug lead.
In my case it turned out to be the throttle potentiometer, and was fixed successfully under warranty. If the harness plugs are clean, this is probably the likely cause. Check that the plug to the throttle pedal is secure, and that the wiring appears OK, but the potentiometer itself (in the throttle pedal assembly) might be faulty.
An injector problem is less likely, but still a possibility.
