The usual culprit for this is dirt roads and the mechanism fills up with dust.  Best solution is get an air hose and blow all around the area.  If you can get a needle end on the hose this will get into the latch area even better.
Do a search on the forum as this topic is something that's come up a number of times before.
I had this happen before I got rid of my Disco.  Got the compressor, wheel spanner, jack handle out of the back shut the door, changed the blown tyre with the spare and then couldn't open the back door to put everything back in.
