Saw Zappa twice at Festival Hall in Brissie. Definitely #1. Another two who spring to mind -- Alvin Lee from Ten Years After & Robert Fripp from King Crimson. Fripp also did a fair bit with Bowie. Also saw Tommy Emm. in the 80's. I had a dance with his missus although I didn't know who she wasI think this list could be mammoth due to individual tastes.
It is hard to compare a Rock player with a Classic music player so I put my list according with my music taste, jazz, gipsy, Spanish and classic.
Django Reinhardt (he was a master of gipsy guitar)
Paco de Lucia
Pepe Romero
Bireli Lagrene
John McLaughlin
Eric Patrick Clapton
George Benson
Andres Segovia
Heck yeah Robert Fripp!
John Petrucci from Dream Theater, featured in the G3 tours with Satriani and Vai. Saw DT live a few years ago.

I reckon there's no other guitarist that you can identify with fewer notes heard than Mark Knopfler. So distinctive.
