Cut all the new shoots at the same time no matter how small they are, and paint/wipe/sponge - not spray - the newly exposed wood with 1 part round up to 1 part water. Only do this if there will be no rain for 24 hrs or you can protect the stump from rain.
Painting will stop any spray drift affecting your new trees.
You need to recut as the old cut will already have been sealed up by the tree, so it is too late to just paint round up on now.
Drilling a cut tree has limited success, it is usually used to kill a tree while it is standing (helps reduce soil erosion) The idea being that the tree continues to take up the poison over a period of time, a cut stump is no longer growing.
Do not worry about round up staying in the ground it does not work that way.
