As far as I am aware, front up to the rego office and pay about $35. Done.
Does anyone know what the process is for getting a home made car trailer registered in South Australia? And where to find the requirements for a car trailer?

Sounds right. I've got a home made camper trailer that was originally built by father in law in Adelaide. To get it registered when I picked it up over there a few years ago, that's all we ended up doing.
To get it registered in WA a few months later, it had to go through a full roadworthy inspection.
