This was a common Defender problem, I had the same issue on my 300Tdi years ago. Sealing it up was only a temporary cure, you have to remove the roof lining to get at the screws that hold the roof to the windscreen frame. One of mine was almost completely undone the others were loose. From memory they were a very large self-tapper that screwed into the frame, two each side.
Problem was caused by too heavy a load on the roofrack as already mentioned. My roofrack alone is close to the limit.
Not sure if the latest Defender is bolted together exactly the same way but if so you may have to remove the roof lining at some point to fix it.
