Going to a 29" diameter tyre will increase yourr evs for a given speed on road, but give you better acceleration and thus should handle hill starts with the van a bit easier.
All, What is the advantage if any or disadvantage to fitting 225/75R16 to a defender ute. The beast (300 TDI) is currently fitted with a disco auto and transfer case. running a 750/16 I'm about to tow a small van around for a while and can pick up a cheap set. I'm not doing any real off road mainly tar and gravel.

Will also put your speedo out... It's a catch 22 - you might appreciate the extra revs on the highway for towing a van, but might hate sitting at 3000rpm all day! From memory 7.50r16's are a 32" tyre, so revs will increase at 100kmh from 2500 (ish) to perhaps 2800 (ish).
You will pay for it at the fuel bowser if you are not towing on the highway. Too many revs IMHO.

It should just do whatever the standard Discovery rpm are if it has Discovery gearboxes and tyres
