If you mean the complete rear axle assembly, quite possible; there will be a few problems but nothing insurmountable. (I assume you mean a 110 Defender - in the unlikely event you mean a 90, very easy). Later Defenders with the Rover diff would be simpler, as the diff centre only could be swapped.
I am not too sure why you would want to unless needing the more robust diff for advanced offroading.
