Once you have solid fuel to the inlet. During cranking you should see a pressure rise in the rail. If the rail pressure doesn't come up. It won't command the injectors in my experience. I have had to change the rail sensor on my d4 when I took the rail out, for no reason at all. It just died.

The LPFP runs, I've pumped several litres of fuel out into a container.
The rail sensor is reporting pressure.
I've cracked injectors and got fuel out, but now that I've done that the rail pressure has dropped from about 350 kPa to 50, which makes me think there is either a blockage or no fuel is being delivered for some reason.
I'm thinking I have it plumbed wrong, even though I've double checked..... But I'm running out of options.
Does anyone have the discovery 3 fuel schematics for EU4, I think the chassis number split is 137A.
I think it's section 310-01C , I bought the workshop manual from AULRO but that section seems to be missing.

I only have the EU3 version. You could always subscribe to TOPIx for a day and get the later one.
The fuel distribution on the EU4 is simpler than the EU3 as nothing goes back to/through the filter.
Tank -> filter -> HPFP.
The return combines the spill from all 6 injectors, and the return from the LPFP via the temperature sensor and through the fuel/oil cooler and back to the tank.
I'm pretty sure the supply & return lines have different sized nipples, so I can't see how you could get it wrong.
Edit: It really sounds like the sort of thing that you see when the PCV isn't closing, but if it was unplugged or there was an electrical issue you'd be seeing faults logged.
350kpa isn't much. If you leave everything connected and your satisfied the HPFP is bled and just crank the engine. How high does it get after 10 seconds. And is it rising?
Note.. The battery will need to be higher then 10v while your cranking it.

Thanks for that bulletman, that has been a great help!
So I've checked everything in the fuel system plumbing, it's spot on, I hot wired the fuel pump to run out for longer and am getting a constant 5 psi.
I scanned again for errors for about the 5th time and got the following, and also a screenshot of the readouts when cranking:
